Do you mean _optical_ and electron microscopy? Or do you mean _scanning electron_ microscopy and _transmission electron_ microscopy?

Optical microscopes use visible light to resolve the image, while electron microscopes use beams of electrons. Electron microscopes can achieve greater resolution (and therefore can resolve images of smaller objects) than optical microscopes can.

Scanning electron microscopes resolve an image by bouncing a beam of electrons off of the target (basically, front lighting). Transmission electron microscopes "shine" a beam of electrons through the target into a detector on the other side (back lighting) to create an electron silhouette.
