Yes you can. Use Miscrosoft's new password protection tool through which you can protect any folder.

If you want the highest security use True Crypt - it's an encrypted storage space protected by password.You drop your files into it and no one can get at them without your permission. Freeware

It depends on what operating system you use but there is free software that can do this job for you if your operating system doesn't.
