your physician's office will give you a stool sample kit which contains nutrients that will keep the salmonella organisms alive until they reach the laboratory. the laboratory will then isolate the salmonella (if present) by selective nutrient agars and will then perform biochemical tests on the organisms they have isolated to obtain an identification.

If you are asking how they test a human for Salmonella, then the answer is with a fecal specimen. There is a kit they give you at the doctor's office to "capture", seal and send a specimen to the lab for testing. I know, because my nephew had to be tested for it.
