The UFT.
(UFT is a collective name for the theories that try to put Quantum Physics and General Relativity together - right now they don't talk to each other mathematically speaking)
You know how there are all those saying about each time you get a new answer it just raises more questions? How about the famous 'The larger the sphere of light, the larger the darkness that surrounds that light' ? Well it won't be true for the first time ever if we get a UFT - we will have hit a limit of knowledge.
So the UFT gets my vote - if we can solve that, then we will have taken a very large step in understanding the universe.
The down side is that it is so complex that we will need another Einstein to solve it.

Viruses, are they living? They have DNA, but no other qualifications to be a living creature. It is hugely debated among scientists who have cam up with a hypothesis of no it is not living, but it has DNA. Viruses do not require food and water two of the basics of life, they get thier "life" from being parasitic, and eventually. Maybe there is a way to live without food and water, maybe our whole view of life is wrong, who knows

For 'exciting' I'd have to say astronomy, because there is so much to be learned and so much happening.
For 'important' I'd have to say the studies of gravity, because it's the one major force we have NO clue about after hundreds of years.

it's a tie between nanotechnology and spelunking(finding the end to some of the worlds biggests caves and finding new ones).

Biotech, because of its applications to human health. Next would have to be space exploration just because it helps us to understand our place in the cosmos.
