Well, many years ago I read a sci fi story about how a cargo had to be delivered to Pluto in a hurry. So they got their top-gun rocket pilot to accelerate at 3g to the halfway point and decelerate at 3g the rest of the way. When he arrived, he was a shuffling incontinent wreck and had to be put in an old people's home.

High positive G's (downwards, like gravity) will cause the blood to drain from your head and you can suffer 'grey-out' or black-out.
High negative G's can cause red-out, which is an oversupply of blood to the brain.
