Don't know where you live, but the first stop is the phone book or to do a web search for "truck driving schools".
Most Truck Driving Schools will require you to come to school with a permit and a DOT Physical exam.
This will require you to go to the Department of Motor Vehicles and study for, then pass the written test.
Then it is off to the Doctor for your physical.
The class(s) will last typically from 1 to 3 months during regular business hours.
The school will USUALLY then take you to Department of Motor Vehicles for your over-the-road exam, including a Pre-Trip, a 4-point air brake inspection and finally the drive test. The school will have covered all of this and more during the classes.
The school WILL NOT, however, act as a placement advisor (like an employment agency). Most schools that are reputable are resources for companies looking for good drivers.
When you graduate, you will be qualified to operate a Heavy Duty vehicle, and will have your CDL.
It will take a couple of years experience to become a truck driver.
Good Luck. This is an honorable trade, and there will continue to be a shortage of GOOD drivers for some time to come, insuring your continued employment.

I have been driving for about a year now and enjoy it. I'd suggest using a recruiting agency to get your first job becuase they work with numerous companies and can find the best job for based on your situation.

I used a company called Truckers Dispatch and was very happy with the job they got me. Their website is http://www.truckersdispatch.com
