On the individual level, it indicates that the speaker is responding to a statement that he believes is superflous or too self-evident to be mentioned.
On a larger social level, the expression "Duh!" signifies at least two things:
1. A decline in the use of sophisticated language--in this case, a phrase such as "Well, I should have thought that were obvious" might have been used in a situation in which a modern speaker will simply say, "Duh!"
2. A decline in general civility, insofar as the expression carries a weight of sarcasm which likely is not nearly so deserved as the ubiquitousness of the phrase would indicate.

It's what one says when the answer is too obvious for you not to know or that you're not that bright to not know the answer. usually it's derogatory.
it can be like the expression "hello!?"

"Duh" used to be the word people associated with really stupid people, so if someone says "DUH!" to you that means that they know the info already and that you are stupid (or slow)
