There is no 100% sure way of knowing someone read your mail. There are certain programs which add a tracking code or web bug. the idea is that when the recipient opens the email, a call is made back to the tracking server and confirms it was opened. Keep in mind "opened" doesn't necessarily mean read.
However, some email programs will now alert you to these bugs or strip them out. Also, some users prefer text emails as opposed to web mail or HTML mail.
You can get a better idea of how these programs work in the article below.

It depends on what email program you use. The email I use a work, I simply click on the email I want to tract and then right click and select history and it shows me. I am not sure if others are the same. At one time and prehaps today, AOL had that option.

Unless it is over an intranet (like in an office) not really. Everyone uses different e-mail clients so Netscape doesn't play with Yahoo to tell Yahoo that it's user read their e-mail.
What some sites do is tell the user they have an e-mail and make them enter a password to retrieve it. Then the website alerts you that they have read it.

Yes, a good way to find out is to call the recipient and quiz him/her about the note.

If you use outlook for mail, you can request a read receipt. If you use another client - search for read receipt in help.

Well, you will usually get a response back. If it didn't go through you will get a mailer daemon or how ever you spell it.
