I love using Mapquest.com to find driving directions, or yahoo.com and click on maps, then driving directions.

It will give you directions the fastest way, plus mileage, and a map. It's better if you type in specific addresses, or else it takes you from and to the center of each town.

Real simple:

Starting in Middletown, take SR 122 across the river through West Middletown. 122 (aka Middletown-Eaton Road) will take you all the way to Eaton. In Eaton, turn right (north) onto US 127. 127 will take you to Greenville - it actually bypasses Greenville, so just watch the signs so that you can exit to Greenville. Altogether, expect the trip to be around 50 miles.

I'd take Ohio highway 127, but I much prefer 2 lane state highways. It's probably the closest route, first of all. I think it may actually run directly through both towns. I'm pretty sure it runs right into Greenville, but if not it runs into 36 and 36 will take you right there. I like little Ohio towns, and it has a few small towns on it. None of them are going to slow you down much. 127 is straight and level. You can go fast on it. There's not much traffic on it unless it's planting time and harvest time. Then the farmers are out on it in force.
Good luck! I like that highway. Good to talk to a fellow Ohioan.
