I use it for my fish immediately, even when they are in the aquarium. They don't mind it. Actually, they swim around in it.
So you really don't need to wait to put your fish in the aquarium after using AquaSafe. I feel like I should tell you this funny story:
Once I put some AquaSafe in a metal container. A week later, I changed the water. I put in the AquaSafe, but it had turned red!
The fish were alright, though. Still, I wouldn't put it in the metal container again.

If you are just setting up a new tank, you really should put in some "Cycle." This is a liquid that has millions of beneficial bacteria in it. It helps speed up the nitrogen cycle that all new tanks must go through before it is completely safe for your fish. AquaSafe is just a dechlorinizing agent with some extra stuff for the fish, it does nothing to help the nitrogen cycle.
If you overcrowd your tank right away, you will most likely lose most of your fish to what is called the "new tank syndrome." What happens is, ammonia builds up quickly because there are no "good" bacteria in the tank yet. Ammonia is the main killer of fish in new tanks.
A new tank should only have 1-2 fish in it for about 2 weeks to give the bacteria a chance to multiply. Then you can add more fish according to the size of your tank. One inch of fish per gallon of water unless you have goldfish and then it is one inch per 3-4 gallons.
You didn't say how big your tank is or what kind of fish you are going to have. Do some research before you add too many fish. Also, do your 25-30% water changes every week to keep your fish healthy.

You have to use it when you change the water, so I would say immediately. I use it on my goldfish, but then again, they are goldfish and they never die.
