Signaling would continue. That is why the neurotransmitters are usually cleared out fast after release.

It may continue to exert an effect on the postsynaptic receptor...

eg. in Selective Serotonin Reuptake Inhibitors - used in depression, they prevent pre-synaptic neuron from taking up the 5-HT
