I wear a 44R jacket/blazer, but was wondering how big a 44L would be on me?

OK, thank you, I thought it meant "large" or something..

If it is too long, couldn't a seamstress hem it up for me?

a 44L is longer than a 44R (the L stands for "long")

When you look at the clothing labels, and see letters, it's talking about length. A S = Short would be for guys 5'7" and under. R = Regular is 5'8" to 5'11". T = Tall for 6' to 6'3". ET = Extra Tall for 6'4"+. Besides being longer in the sleeves, the placement of pockets, buttons, collar, etc... is different to look right on the taller men. Just shortening the sleeves on the jacket wouldn't make it look right on you. When the jacket is buttoned, you might end up with the V ending, not in the middle of your chest, but closer to your stomach. It won't look as nice. If you want a lower or higher opening on a jacket, then buy a style with the correct amount of buttons. The more buttons, the higher up the jacket will close.
If you are 5'11" and feel like a R is too short, go for the T. My suggestion would be to go to a men's suit store, like a Men's Warehouse if you've got one, and talk to the salesperson and the in-house tailor. They can help you find the right fit and style.

The difference is in the length of the jacket and it's sleeves, as well as the inseam in the pants. So, yes, it does make a difference. You would probably not want the L unless you were a minimum of 6'2".
