Durring normal freeway driving at 55-65 the engine speed is normal durring acceleration. Soon after I reach cruising speed, the engine speed increases. If I push slightly on the gas pedal the engine speed decreases and I can feel the tranny "lock in".

Also, if while cruising, I take my foot slightly off gas pedal it tends to "lock in".
Trans fluid fairly clean. Not burned.
The transmission works wonderfuly all other times.

What gives? I don't want to waste a bunch of money on the wrong repair.

2500 series truck probably has a4L80 or 4L60.. torque converter locks in at about the speed your saying you experience rpm drop a scan tool will tell you if there is a slippage problem Only way to diagnois it right

It never hurts to have the transmission fluid changed.
Once I had an AAMCO transmission shop tell me that my tranny was going out on my Trans Am because it wouldn't shift. After they had dropped the pan and changed the fluid it worked fine. They looked pretty stupid trying to cheat me like that. I'm not a transmission expert but at least you won't be wasting money as it's a good maintenance thing anyway.

Two suggestions. Number one, put a quart of Lucas tranny treatment in with your tranny fluid. Try that for a couple of days. That stuff can work miracles!!! If that doesnt work, take it to a tranny shop, call around to find one that will look at it for free. A good idea would to ask around for a trustworthy tranny shop while you are at the parts store buying the Lucas. Good luck.

Sounds like maybe a belt might be slightly slipping. I would first try some lucas tranmission hunny and see what the does for you. It fairly cheep around 10 to 12 bucks depending on where you get it.

When is the last time you had the trans serviced? Good chance the filter is plugged and cannot get enough fluid through.

You need to have that thing hooked up to a diagnostic computer [or whatever you wish to call it] because your shifting would be almost completely electronic.
