Manual.Viruses usally attack system32 folder.You can sort files in this folder by date with Windows explorer and manualy delete any recent exe, bat, dll or other files that you think it's virus.Also you have to remove them from registry. But if you don't have expirience with this don't do this.

Anti-virus software is essential. I am using Norton anti-virus, and it's not free. You also need software to scan for spyware, etc.
