Here is a decent source attached below. There are lots of sites on the internet. Spend a lot of time on each cover letter, customizing each for the employer and the position. Tell the employer what the need to know: Why do you think you would be a great match for the job. Proofread the heck out of it, and if you are not too self-conscious about it, get a friend to review it.

You should be able to read the cover letter out loud and it should sound like you are talking. It should be easy to read and it should provide value to the intended audience.

Let's say I am the hiring manager. If I am looking through a stack of cover letters, I am going to be bleary-eyed and anxious about the task at hand. In that frame of consciousness, I do better if a candidate just gets to the point. Bullets are great, white space is great, succinct sentences great.

I won't judge based on "Regards" or Sincerely" in the closing, I don't care about enclosure notation--unless I am hiring a secretary. HOWEVER, misspelled words and awkward grammar will jump out at me.

Don't mess with fancy paper or a special font unless you are going into a field that expects immense creativity. It can only hurt you. Think about it, "Mike, why did you choose that person?" "Well, I may have made a mistake, but they seemed like the perfect person. I really loved the high percentage of cotton in their resume and cover letter, and the aquamarine hue? It was just incredible. It just made his resume stand out from the others!"

Sorry, but another word about making your cover letter and resume stand out, if it concerns you like it does me... In most cases, an employer wants a qualified candidate who fits into the job and poses very little risk of making the hire look like a mistake. The best way to stand out is to network with the people you know, so that one of them can provide your resume to someone they know and say, "Hi, Pat, how's the family? Great party last week, I hope you've recovered. Say, I know you are looking for a person to fill an open position. Here's a resume from a friend of mine. I've known her for x years. I ask her for her opinions once in a while, as she is an expert in xxxx." Truly the best way to get a job.
