It depends. It could be a guy who has lots of girlfriends and he is talking about his favorite, it could mean the best friend of a girl (gals call their female friends girlfriends, and it isn't a sexual thing AT ALL), it could mean a guy who has a lot of female friends and he is referring to his best friend who's a girl, or I suppose that in very rare instances it could refer to a lesbian with many girlfriends. But I've never heard it said in that way before. USUALLY, it means, and I would assume it does in your case too, a girl talking about her best female friend.

That question is hard to answer because it is taken out of context. What are the circumstances of the question? Is it a question about a female/male relationship OR a question about a female/female nonsexual relationship OR what exactly? That's like asking "What is the last day"...last day to what? To a girl, a best girlfriend is someone who you respect and trust enough to share with her all your secret grudges, crushes, problems...all that you would never dream of telling your mother.
