Well, other than the usual, ah, enlarged presence, I would guess that you could tell Mrs. Mouse is pregnant if she has started knitting little booties for her kits to wear while still in the litter.

Gestation is in about 21 days, so you'll know for sure in less than 3 weeks.
Usually they do get big, especially near the end of pregnancy. If it's early on, you don't have an easy way to know.
If the female is old enough to breed (about 5-7 weeks) and she was in with males, odds are she was bred. You can confirm a recently bred mouse by looking at her vaginal opening for a mucous plug.

Mice are only pregnant for about three weeks before giving birth to litters of 4 to 12. Their little tummies will begin to distend with the first week. After week, two it will be very apparent. As soon as she gives birth, she can be re-impregnated immediately.
