You can't start knowing for sure until the sex
organs sprout. Look very closely all over the plant
where the stems connect to the main stem. The females
will develop tiny little hairlike strands in the shape a a V.
The males develop little pods. By the time you identify a
male it is often too late (assuming you don't want it ruining
your females by fertilizing them). Keep track daily.

Most Anime characters don't cross-dress, so what you see is what you get, (meaning that if the picture looks like a female, then the character is most likely a female.) However, there are some characters that are known as "traps", (these characters are actually males with feminine traits that occasionally cross-dress.) There really isn't anyway of knowing whether these charaters are male or female by looking at them, (the only way to know is to watch the Anime in which they appear and find that out yourself; it's nearly impossible to tell whether a character that you've never seen before is a trap or not.)

The males almost always show their sexual characteristics first. In the beginning (they're called pre-flowers), they look very similar and it's very hard to distinguish the difference but if you wait a few more days it becomes obvious. The boys' flowers grow into clusters that look like little bunches of grapes. These clusters take time to grow and mature (swell up) before they will open and release pollen. The girls look very different, sporting telltale fuzzy, white hairs (stigmas) that take longer to form the clusters.

No. I can't even tell the difference when they are mature. Of course, I have never even tried because I never cared.
