I am in a network of about 10 people, yet my signal is listed as low or good, at 48 to 54mbs. It is a linksys router, and I have a standard basic wireless internet card, how do I strengthen my signal, and prevent loss of connection?

The router is positioned on the 2nd Floor, on top of a doorway. My computer is on the first floor, at a down diagonal angle at about 30-35 feet from the router. There are 8 users on the 2nd floor, and only two on the first.

If your card supports has an external antenna then you can get better reception by replacing the standard antenna with a high gain antenna. Linksys, Belkin and many others sell these.
Or you could upgrade to a MIMO-capable card for better range and reliability.

are you in the same room as the location of the wireless AP/router? if so, then make sure you have a direct line of sight to the antenna of the wireless AP. You can even consider buying a small antenna to connect to your wireless internet card (there should be a small plug on the card for you to connect your new antenna to, try to buy a directional antenna rather than an omni and then point the antenna towards the wireless AP)
If you are not in the same room as the location of the wireless AP/router, then it makes life a little bit more difficult, 2 methods, buy a bridge AP, or buy a pair of "power LAN" which essentially uses your electricity socket and lines to extend your LAN. The former can sometimes be a pain to setup, the latter is just expensive.
Third scenario, and if you are allow to change the linksys or allow/afford to buy a new one, then buy one of the MIMO type.
