Change your medium of writing. If you're typing, get a pen and paper and start writing. If you're writing with pen and paper, start writing. If both fails, get a voice recorder and start talking.
There are different kinds of Writer's Block.
One is the Ideas Block. That's where you run out of ideas to write about or to expand upon a certain topic or theme.
Another is the Words Block. This is where you have all the ideas in your head but you don't know the best way to express it in words.
There are other kinds of blocks but I am myself stuck in a block to remember what they are.
Okay, to solve the Ideas block, simply ask yourself a few questions concerning the topic at hand. Ask more Why or How or What or Who questions. Get into as much detail as possible.
If you have a Words Block, simply state everything you have in pure simple terms. Get blunt. Write lousy. Even if you have to write sentences like, "Peter is a boy". The idea is to keep doing this until your brain starts to spurn out all the good stuff hidden inside your head. If you're still not satisfied, go ahead and plagiarise another author's work and rephrase, reword, restructure his or her words until your reworked one is significantly distinguishable from his or hers.
Or maybe you're just being too much of a perfectionist. Stop being a perfectionist.

Start by reading other material for ideas.
Write an outline of plots or points you would like to make.
Write about your characters, the theme of your story, what kind of atmosphere do you want your readers to experience?
How are you going to write your text? Is it going to be in the past or present?
Will you let your readers know what your characters are thinking or will the readers have to discover them through dialog?
If you are writing non-fiction, try interviewing someone with an interesting job.
It is extremely important to take a closer look at your environment and try to notice what others don't.
Or you can go back to basics and brainstorm by just writing whatever is on your mind on a sheet of paper.
If you don't want your work to seem like someone else just try to write down the world through your eyes. It's not easy because there are somethings you may not like to mention in your life but sometimes that makes your writing original.

I call those moments "non-inspired", as opposed to my "inspired" ones when I have a real rush of creativity. I try to write through them -- at least capturing the ideas that I've had but haven't really fleshed out yet. You can always go back when you're feeling more of the flush of creativity upon you and elaborate, expand upon, or dress up what you've written. But at least you are doing SOMETHING productive and not wallowing in your frustration while you wait for it to pass. The outline suggestion someone offered is a good one. Now, if you're feeling completely dried out and utterly devoid of ideas, that's a little harder. In that case, I'd just go and start researching something I have a passing interest in and see if anything takes root.

I've read that you should sit down and write, no matter what. Even if it's junky and you'll have to throw it away. Eventually the right words will flow.

It depends. If you're having trouble in the middle of a piece, work on something else. If you're having trouble coming up with any ideas, read something, prefferably something you wrote.
