You CAN'T TELL if a lady is virgin or not from intercourse. Nor from anything else, really; there aren't any tests that can be run for "virginity."
There are some signs of virginity, like bleeding caused by the hymen tearing, but they're not a guarantee--some women lose their hymen through exercise or doctor's exams, and some are so flexible and stretchy inside that there's almost no pain, and no tearing or bleeding.
First time sex is usually painful for the woman, but with a careful partner, the pain is over quickly and the fun part is what she remembers more. It takes going slowly, making sure there's enough lubrication, and being very still after penetration to give her a chance to adjust.

Well, first of all they wouldn't be a "lady" unless you're into 40 year old virgins. Second, all you have to do is check to see if the hymen hasn't broken. If you don't know what a hymen is then you need a little sex ed prior to venturing into these unknown territories.

Usually on the first time there is blood. No blood means not a virgin.

Sex for the first few times is painful. After that it's all good. unless you manage to get yourself into interesting positions. sometimes that can hurt. has for the other question, i can't answer that.

You can't.
Tightness, blood, all that doesn't mean anything.
Not all women have a hymen intact since it can easily break by doing excercise or horseback riding.
You have to ask her and trust her answer.
