As a born and raised San Franciscan who's traveled America and the world pretty extensively, I'm going to back up Sarah and say - San Francisco.
That's *not* just because I was born there. There's a sense of community in The City that's unmatched anywhere else in America.
You didn't ask, but I've found Tokyo to be the friendliest city in the world. It amazes me that the center of a megalopolis of 27 million people can still be so polite, friendly, and virtually crime free.
And Mortiferia, Portland... I own a business there and I strongly feel that it has a loooong way to go. It has a few pockets of greatness, but sadly, that's about it.

Charleston SC you would not believe how friendly it is. Everyone there is speaks and in the stores even the staff is nice. At the hotels all the help is very nice and actually helpful. It was voted the friendliest city to visit.

Technically they don't consider themselves "American", but puertoricans are known to be the happiest people in the world. They live in a lovely little island, have amazing weather year round, and will even celebrate hurricanes as if they were New Year's Eve! It's the friendliest place I've ever been in, just don't make the mistake of calling a puertorican "americano"...

Houston, TX. I was there on vacation visiting some relatives. They are so nice and friendly. They will just talk to you and say hi on the streets. They don't cut you off in traffic and if they accidentally do they don't swear at you. If you accidentally cut them off they don't flip you off. I was shocked when I got there because I live near Pittsburgh and everyone is rude here. I didn't know what to do when they were nice and talked to me. No doubt about it, Houston has to be the friendliest city in America.

Ever hear of southern hospitality.
We went on a vacation to Texas. Everyone was so friendly
took time to chat.
Young men in Killeen held the door for me and older women.
We had a wonderful vacation.
I would say Killeen Texas, it is a military town of young people who were so well mannered.

Its got to Atlanta. Southern charm in a big city, it was the most pleasant big City I've seen.
If small town's count its Hope Arkansas, a slice of the good life, if you've ever been there you know what I mean.

Portland, Oregon. I live there and compared to any other city I've been to, it's the friendliest and most open.
Don't mind us damn hippies, we don't bite... promise! That would probably be harmful to the earth or something, for one thing. ^_~

San Francisco is like the BEST city in America....I have been to them all...LA, New York, Chicago, Detroit, Philly, DC, Columbus, and even out of the US.....San Francisco is the nicest, cleanest, and most polite city....the only downside? Way too expensive to live in :(
