Hm. Good question. You could try attaching a link to a small MIDI in your .sig, or you could try adding musical notes (C-C-C-C, E, D-D, C-C, B-C, or somesuch), or ASCII art. Unfortunately, technology is limited in this regard, so you may have to settle for lesser, here.
