"Yolk" is singular so you use "is".
Added later: Obviously I wasn't focused on the "white" part of the statement. If this was intended to be a grammar question, my answer is okay. If it was intended to be a factual question, obviously neither statement is correct, at least if we are talking about standard chicken eggs. There may be some kind of eggs that have white yolks! At any rate, since this is under the classification Polls & Surveys, it's difficult to assesses exactly what the intent of the questioner really was.

The correct verb to use would be is. If you diagram the sentence the following parts of speech would be correct grammatically::
The- -is an article
yolk - singular noun and the subject of the sentence
"of the egg" - is the prepositional phrase identifying the type of yolk
is -verb, singular conjugation used for singular subject
white -adjective describing the yolk.
After identifying the parts of speech when you diagram sentence it is easier to determine the correct grammatical usage of the english language. Even if the grammar of the sentence and the punctuation are correct it does not mean that the content of the sentence is accurate. Egg yolks are not white, they are yellow.
