Go to Stanley Park. View the scenery and the totem poles. Visit the aquarium in this park as well as the zoo. The zoo has some rides for smaller children.
You could let them ride the sea bus (hovercraft) from downtown Vancouver to the market place on the shore of North Vancouver. It has a lot of little shops and food vendors who sell a variety of ethnic food.
Granville Island near downtown is worth a visit, although I am not sure how much it would have for toddlers.
The Univeristy of British Columbia, situated in Vancouver, has a Museum of Anthropology that is said to be fantastic. I have never seen the museum myself. The new library also is worth a look.
Capilano Suspension Bridge over Capilano Canyon in North Vancouver is a shaky walk but offers an interesting view. A trip up Grouse Mountain is also worthwhile sightseeing.

If you're looking to stay close to the west end then for sure do Stanely Park. There is a water park for the little ones, the petting zoo would be great for them too. The sea wall surrounds the park and you could walk part of it (all of it would be a bit much for the kids as it is 10 kms) Robson street is an expensive place to shop but it is right in the west end. China town is not too far and there are lots of neat stores and great food there. Granville island also has great stores and a great vegie market and the food is fabulous. You can catch a little ferry by the aquatic center to go to GIsland. Science worls might be fun for the kids but if there are under 5 it probably isn't worth the $$$ although the imax theatre is pretty cool for adult too, you may wanna just tken in a movie there instead.

Vancouver is awesome.
Try Stanley Park. Your kids will love it since their is a petting zoo, aquarium, small water park, picnic areas, etc.
Robson Street would probably be a very nice place for you and your wife to go for great shopping.
Also, check out English Bay if you are there on a Saturday or Wednesday night for the "Celebration of Lights" are on right now. Thats where different countries come to compete in a fireworks show.
Granville Island is also amazing. Lots of little stores and street performers.
There is so much to do in vancouver.

on the North Shore there is Lynn Valley canyon, in Richmond there is Steveston with all the fishing boats and people flying kites, right in Vancouver is also Queen Elizabeth park were there is a great hill the kids can roll down, Granville Island has one building with just things for children. Second beach in Stanley Park has a wonderful pool perfect for lil people and adults....have fun and come back soon!

My wife and I were in that area one year ago this month. This is what we did .
Explore the city -- downtown is great. Great park called Stanley Park that is beautiful for families. Also the ferry ride over to Victoria is very nice indeed. Finally a drive up to Whistler is very beautiful -- Whistler is getting ready for the 2010 Winter Olympics and it is very nice. Lots of family oriented stuff there as well and a gorgeous drive.

Go to science world for the kids. Take the tram to the top of Grouse Mountain for dinner. Storyeum in Gastown is interesting. Go shopping and eat on Robson Street. Check out the aquarium in Stanley Park.

best thing to do with kids is go to Stanley Park - kids need to have tactile activities - this means hands on with nature - throwing rocks in to the ocean... seeing the birds in the lagoon etc.. walking
