CoT curves and the CoT1/2 describe the time it takes for homologous sequences to re-anneal, which is a reflection of their relative abundance. Highly repetitive DNA sequences have many copies per genome therefore, if you melt out all the DNA duplexes and watch for re-annealing by the change in UV absorbance over time, repeated sequences will re-anneal very quickly, giving you one rapid series of deflections in the plotted CoT curve. Single copy genes will take much longer to find their complementary strand and will thus take much longer to create the necessary change in absorbance indicative of their having become double stranded again.
