Tough questions as they're not really in the same class of cars. True both are considered "entry level luxury/sports sedans" but the similarities end there.
Overall, the IS350 is a superior car: rear wheel drive, bigger, more powerful engine, more modern design. However, to get this, you're going to pay many thousands more for it than a similarly equipped TL. If you have the cash, go for the IS350.
If you're looking for a new car, a significant competitor to the IS350 will be coming out in the next few months: 2007 Infiniti G35 sedan. I would give that car serious consideration too.
