It's a feeling you get. Sometimes it takes awhile, because people are always SO into their own little life events, or some major life event they won't discuss, but is really bothering them, so they aren't themselves at the time. I'm sure you've gone through that. I've found that if you are yourself, be it bad or good, people will respond to you honestly. If you are like a manager, and put on an act of superiority, like you are better than your employees, then you'll most likely get false niceness from the employees, because they need their jobs. Lots of managers take advantage of the position, just to feed their own ego, or get sex. But basically to answer your question, you can feel it, with time.

consistency is one thing one can look for. The verbal, non-verbal, looks and any other aspect of the personality will corroborate in case of an honest person where as there will be some inconsistency some where in a dishonest person,which can be perceived when you interact with him; however nicely he may try to conceal it.The more prolonged the interaction, the higher is the chance that you would find out this inconsistency in a dishonest person.

By first being honest from deep within, for a good length of time, and getting used to it, knowing how it feels, then looking at another person, very very easy, because, by the our perception level deepens so good, we can almost smell it apart !

Simple method is , when honest man speaks he will always look into your eyes, dishonest man never speaks that way.
They always avoid direct eye contact!!

Pay attention. You will know. Unless they are habitual liars. You have to know the person to know sometimes.
As a general rule, if it sounds too good to be true, It usually is.

dishonest people will try not to look in your eyes when they are talking, they may fidget or seem distracted.
