The diamond museum's are fun as was the torture museum. I enjoyed the cannabis museum as well. You should check out the floating flower market. Some really nice shops etc in that general vacinity as well. And finish off in Prix Damis. A really nice coffee house not too far from Centraal station. Plenty of good maps are available when you get there and the tourist info opposite centraal station has loads of ideas. It's a fab place and you'll have a blast.

For nightlife try any bars in the Leidensplein ('scuse the spelling!) The atmosphere is fantastic, and not at all scuzzy (the red light district is really rough at night!) Anyway, Leidenspein is this cool square in the heart of amsterdam, with plenty of Irish bars, sports bars, etc, a good time will be had I promise...
In the day Anne Frank Haus is pretty amazing, and the VanGough museum is nearby.

I think it is just fun to go into town in the evening and watch the tourists do stupid and wierd things. I was there last week and we saw a drunk/stoned tourist harassing restaurant patrons and then get tackled and brutalized by all the waiters in the area.
The Reichsmuseum is good though too. Nice Rembrants and good Dutch realism.

Just walk around, hire a bike and just tour around the city. The obvious is well-known, its the little things you will notice on your tour on a bike that will make your stay unforgettable!

Definitely walk through the red-light district. It is one of the most surreal things I ever saw. Go at night on a weekend night - it is super crowded, but safe.
