In 1922, Ted Healy, a boyhood friend of Moe and Shemp Horwitz started the 3 Stooges. Moe and Shemp changed their name from Horwitz to Howard.
In 1925 the trio spoted Larry Fine (Louis Feinberg in real name)
For years there was 4 Stooges but they were known as Healy andHis Racketeers.
In 1931 Shemp quit and was replaced by his younger brotherJerry. He had full head of hair and a handsome mustache but Healy insisted that he shave them both so he became Curly.
From 1930 to 1950 the Stooges had four personnel changes. In 1946 Curly suffered stroke and retired; Shemp returned to Stooges until his death in 1955, he in turn was replaced by Joe Besser (Joe) and Joe De Rita (Curly Joe).
So originally there were 6 Stooges.

Moe Howard and Larry Fine were always a part of the act. Though Moe's older brother Shemp was in the original stage act, he left before they stared doing shorts, and that was when his famous younger brother Jerome "Curly" Howard joined the team.
While filming the short "Half Wits Holiday" in 1946, Curly dropped over with a stroke. Even though he lived on for six years afterwards, his acting career ended that day (outside of a brief cameo appearance in "Hold That Lion" the only time the three Howitz (staged named Howard) brothers were seen together in the same film). Shemp came back to the team at that time (1946).
Curly's health grew continually worse as he was confined to a home. He died at the young age of 48 in 1952. Shemp was the third stooge from 1946 until his unexpected death of a heart attack while returning in a cab from a boxing match in 1955. He was 60.
When Shemp died, they needed to find a third stooge to finish out their contract. Auditions were held, and, even though Moe wanted Joe DeRita, Colombia Pictures chose Joe Besser, since he was already under contract with them. When the president of Columbia pictures had died, the studio cancled the Stooges shorts. Joe Besser's wife took ill, which nixed a tour the three were planning on going on.
When the movies came a calling, Moe was able to get Joe DeRita in as the third stooge. After having made three movies, they started going on tour, until Larry Fine had a stroke in 1974, and then died in January, 1975. Moe soon passed on in May of the same year. Joe DeRita lived on until Independence Day, 1993.
So, to answer your question, six different people have played roles in the Three Stooges. Moe Howard and Larry Fine were with the team from its start to end, and the other four were the third stooge, until either their health took a turn for the worst, they died unexpectedly, or had other personal matters that prohibited them from continuing. Joe DeRita, the last stooge to play the third stooge, and also the last to die, was once quoted as saying that being allowed to be a part of the legendary team was, "The greatest thing that had ever happened to him."

There were orginally 3 stooges hence the name Larry Moe & Shemp. Moe & Shemp's brother Curly took over for Shemp because he wanted out. Then Curly died Shemp came back. he died and then came the Bad Joes. Larry died in the '70s and about 6 months later so did Moe. Kinda said they were friends to the end.

Always thought there were six overall. Larry, Moe, and Curly were the original three...then Shemp took over when Curly died...then Shemp died and Joe Besser took over as "Joe"...then he quit and Joe DeRita took over and was billed as "Curly Joe", since he resembled the original Curly.
