honey you have given two situations. let us analyze them one by one.

falling in love with your best friend is the sweetest thing on earth. if your best friend becomes your lifepartner, life becomes heaven. because your best friend understands you most. there is very little scope of misunderstanding. so there is NO reason to prevent falling in love with your bestfriend.

BUT IF, she/he has already a boyfriend/girlfriend, then the answer is NO. then you'll create only misunderstanding between them, and a friend should never do that to a friend, a bestfriend, right? but as that person is your bestfriend, so you can always talk to her/him and clarify things, who knows? situation may change? or may be your friendship may become more deep.

I wouldn't prevent myself from falling in love with my best friend of the opposite sex who knows he might be the one.

I think you should prevent it.
I fell inlove with my bestfriend but he did not have a girlfriend.
When the relationship went wrong I lost my bestfriend.

You cannot prevent this. Sooner or later this will happen.
As an advice: Don't go making friends of opposite sex. :)

It probably wont work out, but dont waste the TIME you have now to do it.

She could hate you one day for farting or something, and you'll regret not taking advantage of it.

i think u should not prevent urself to fall in love with ur opposite sex friends. it is ur own wish whether u want to love him/her or not.
it does not matter whether ur opposite friend has already a boy or girl friend. may be he/she likes u too. he/she may like ur friendship.
so it is ur own wish in this matter.

some time you have to take a chance... life can get dull with out taking any risks... But in your case I would wait at least until she is single again to tell her how you feel...
