Yell and holler, tell them you were treated unfairly by the doorman, or parking attendant. Threaten to go stay at the Four Queens. Complain about how yucky it is there. Demand a comp.
Or maybe you could just ask real nicely. Tell them you will do all your gambling there, and actually do it. Casinos have what they call comps. Usually you have to be a regular, or gamble and lose a lot before you get one.

If you haven't already done this when you make the reservation, then ask them if they have any special room rates running. Also, ask if you can apply the discount to an upgraded room.
If there is time, go online to the website and register for their player's card. You may get a special deal just for signing up. Some of their slot at table game tounaments have special room rates attached.
Playing dumb sometimes helps. Tell them you didn't know if they tell you that you should have asked when you reserved.
Try telling them that this trip is for a very special event, see if they volunteer a special room for you.
I can't speak for Fitzgeralds in Vegas, but I find the service at their Reno location to be very friendly and understanding. Maybe mention that, as if to say you may reconsider returning to Vegas and going back to Reno instead.

Ask nicely... you don't want to come off rude or "entitled" or you won't get anywhere with the host. If you are a member of a frequent stayer program, then mention that, if not just ask if there is any possibility of an upgrade. Some hotels have more availability and can, others have tight restrictions. Just remember that you kill more flies with honey than vinegar... You may also call ahead and mention if it's a romantic getaway or a birthday or any special event. If they have that on file it could help your chances (just don't lie... thats lame) remember the hotel is only a part of the experiance.

Try calling the hotel directly before you go. Ask nicely. Like someone else said - mention you do all your gambling there.
If you do it at check in, you might not get it because often rooms are assigned by computers the day before based on who's checking out.
Another thing to try is call the hotel and ask for the name and fax number to the front desk manager. Send that person a fax asking for the upgrade - especially if it's your first time to that hotel and/or it's a special occasion. The worst they can tell you is they can't do it. (In which case, you'd probably get something else in return - a dinner, show tickets, or at the very least, a fruit basket.) I did this at the Luxor in 1999 when I took my mom for her 50th birthday. We got a jacuzzi suite for a weekend at the weekday regular room rate. (A $259/night room for $53/night!)

you can ask...but chances are they will want money. Or a good way to get a upgrade or even a free room is gamble a lot of money in front of the pit boss and then just ask them. Sometimes they will hook you up.

Get a players card as soon as you get there and ask to meet an executive slot host and they will tell you the best way to get comps and upgrades.
