The Luxor is awesome, Circus Circus is nice...>DO NOT stay at the Frontier.....its dirty and gross!

The Wynn in Las Vegas is the new "best" place -- but pricey. You have to at least see it.
Belagio is a close second. Then Venetian. Then Mandalay Bay very close.
Then there are a bunch -- my favorite is still the Mirage. Not too pricey, very nice and clean, nice spa, good casino, shows, stuff to do, close to everything else.
I think Ceasar's is overrated for the money. Same with Treasure Island, Luxor, Monte Carlo (is a wannabee elegant but easy to see through it) etc.
There is a big drop-off IMO to the places like Circus Circus and Sahara. Then the near-seedy places like Flamingo, Harrah's, Stardust, Riviera etc.
Downtown has the Golden Nugget -- very nice for the money (maybe the best nice hotel price-wise). But it's downtown so you have that to deal with.
The Four Seasons is a non-gambling hotel that's very nice. The key is you don't have that casion thing happening all the time you're there and you can go to casions but not having to stay at a casino. It's very nice -- very pricey.

Wynn Las Vegas, is by far the best hotel in Vegas. You didn't qualify your question as to best for price, etc. If money is no object, and are just wondering what is the best hotel in Vegas, the the answer has to be Wynn.
If you are just wanting to go check out some places, Mandalay Bay, CP, Mirage, Bellagio, and Venetian are all worth seeing as well.

An amibiguous question, since it all depends on what you mean by "best." Some of the off-strip hotels are cheap...like $25/night. The closer to the strip you get the more costly they become. As for which is "Best" that depends on what you want--a pool? Great room conveniences (fridge, safe, kitchenette)? Easy access to shows? Spashy elegance? My own favorite (based on cost considerations) is "Circus, Circus" which is low-end with lots of room amenities, as well as central on the Strip. Most hotels offer lower rates from Sunday thru Thursday nights. If cost is a factor, select these nights, no matter which hotel you choose.

The Wynn hotel is beautiful, I plan to stay there next time. I stayed at South Coast last time and it was great because it was brand new. Treasure Island is cool, the Mirage will be good once they finish remodeling. Circus Circus is crap, dirty, gross. So is the Stratosphere, it has an odor.

WYNN- luxury from floor to ceiling
BELLAGIO- italian renaissance
Caesar-- but insist on the new tower- they have flat screens in every room
Good stays-- fun places
Luxor
Westin
Paris
Aladin
Cheap

stratosphere
Off the strip and lovely
JW MARRIOTT in summerlin 10 miles form strip
RED ROCK in summerlin 10 miles from strip
Southcoast--south of the strip 5 miles

Best Meaning Cost or Luxury?
THE hotel is really nice ( LCD tv's over the bath tubs)
Cost wise I like the Stratosphere Cheap rates.

I liked Terrible's Hotel and Casino. Good price, decent location, good food, and comfortable beds.
