Do you mean using a sewing machine? If you mean using a machine, then you're probably going to need someone to show you since this is a very basic question.

If you've threaded the machine, lift the presser foot and put the fabric under it. The fabric will be pulled under the presser foot AWAY from you -- I don't know if you'd consider that horizontal or vertical. So if you're sewing a seam, put the bulk of the material on the left so that only 5/8" of fabric (the seam line) is on the right.

If you're sewing by hand, hold the fabric so that you're sewing horizontally.

If you're a beginner sewer and are just starting to use a sewing machine, I suggest you take a class to get you started. It's very hard to learn from web sites or books. Most stores that sell machines also hold classes; also, many schools or rec departments offer beginning sewing classes in the evenings. And check local fabric stores to see if they have classes or know of someone teaching classes.

Good luck! I've been sewing all my life and I've found it a really rewarding hobby -- but it does take some time to learn the basics.
