Well it can be as easy as looking at them or it could be impossible to tell the difference.
There are five types of guys
Flamers
Gays
Undecided
Metros
Straights
Flamers are easily spotted. Their emotions are displayed to the world and they love to be in the spotlight
Gays can be difficult sometimes but if you just watch their behavior around the same sex and opposite sex you can kinda get an idea. They tend to be more eccentric with women and try to display their feminine side with women while maintaining more of a masculine side with men.
Undecided are usually very willing to experiment with both in order to figure out what they really want. Not to be confused with Bi's which I don't believe exist in men. They like one or the other more.
Metros can easily be confused as gay but typically have a keen sense of the feminine side but do not prefer men. They also have the advantage of attracting women like gay guys do but metros get to follow through. haha
Straights are your typical straight man who can be smooth with women sometimes but openly displays their distaste for men. Some are sexually confident in their orientation that they can be confused as gay but they are really not.
If you do decide to blatantly ask, their response can tell you a lot about them.
Punch in the face: could be gay but probably not
"Why would you think I'm gay?": closet case
"No, dude. And F U": Not gay
"Of course": Duh, gay
They begin to flirt but stop: Humorous straight man
Flirt and don't stop: Gay man
Hope this helps and remember, their orientation does not define the personality. I am good friends with both gay and straight. Just make sure they know your orientation and you can get along with anyone.

pink clothes
lisp
falsetto voice
general aloofness

Well, you could ask outright, but that's rather rude and pretty personal, don't you think? The real question, though, is why should it matter to you? Unless you're interested in him as a potential BF, it shouldn't matter. At all.
