I'll agree with the person that said Yakitate! Japan!
Here's the ANN summary
zuma Kazuma isn't terribly clever, but he's got a good heart and great skill - at baking. Since childhood, he's been on a quest to create the perfect bread to represent Japan internationally. Now, he seeks to enter the famous bakery Pantasia, in hopes of reaching his goal. But plots abound...

She asks for any aliens, time travelers, sliders or espers to join her. Watching her weird behaviour is Kyon a boy who sits in front of Haruhi and is the only person who talks to her. Commenting on Haruhi's joining every club in school and then quitting Kyon unwittingly gives Haruhi an idea to start her own after school club. Chocola Meilleure is a happy-going and optimistic girl. Together with her friend-***-rival, Vanilla Myu, they are potential candidates to become Queen of the Demon Kingdom. After obtaining their magic wands, they set off to the human world to gather the hearts of unsuspecting humans. Whoever gets the most hearts will be the crowned Queen. The hearts reflect the feelings of each and every individual and as a result, these hearts have different colors. The value of each heart will be dependent on its color and based on the value of the hearts they have collected, the candidates can buy the goods that they want back in the Demon Kingdom. As far as DVD's you might want to try
Kyou Kara Maoh?!
Yuri Shibuya was living a pretty normal life. That changed the day he was dunked into a toilet after an attempt to save a classmate from a gang of bullies. Instead of just getting a good soaking, he's pulled in. The next thing he knows, he's in a world that vaguely resembles medieval Europe. If that's not odd enough, he's told that he is to be the next Maoh, just because he has black hair and black eyes. The Maoh is the King of the Mazoku, who are coexisting not-so-peacefully with the humans in this world. Much to his subject's dismay, he's totally different from the rulers they're accustomed to. He's kind, considerate, a believer in justice, and not willing to use violence to solve conflicts. Not exactly someone they want running a country on the very brink of war. Now, Yuri has to deal with trying to become a good Maoh, while at the same time attempting to adapt to this land's customs and culture, all in a world where the tension between the humans and Mazoku is reaching its peak.
Haunted Junction.
Saito High is a high school with its spiritual beings and a principal whose hobby is collecting occult paraphernalia. The school has the prophecy-chosen members of the Holy Student Council to protect it. These students have problems of their own; Haruto Houjo, the president, has a way of summoning spirits, and would resign from his duty for a normal life if it weren't for the principal. Kazumi Ryodo, the next member, is good at being possessed and has something for the Hanako. The last member is Mutsuki Asahina, who is a master of exorcism and is attracted to small boys. Unfortunately, these three are in charge of keeping the school in line and defending the world from supernatural beings, and this mission brings them through wacky predicaments.
These DVD's are my favorite one's.
I'm sure you'll hear about pleanty of the Mainstream ones from other people
Fruits Basket
Dragon Half
Gravitation
Abenobashi Magical Shopping Arcade
Azumanga Diaoh
Plastic Little
These are all good comedies too, but you'll be able to find the informaition on them fairly easilly if not at the animenewsnetwork site.
Good Luck!

Yakitate
Its an anime about baking bread, you can watch all the episodes on Youtube. Its very funny becuase they make bread baking seem very serious often having silly competitions with it. And they Often poke fun at other animes. Ive seen Naruto, FMA, DBZ, Sailor Moon, Princess TuTu, and Akira made fun of.

Not a series... but more like a mini-series... SamuraiX. The 2 DVD set (Betrayal and something else... I forget). Not comedy for sure. If you can get your hands on it.. and like old school stuff, get Doraimon.
