Off of what? Nail polish remover should take it off anything!

Depends on what it is on. You can always try nailpolish remover, but remember -- if it is on cloth, it may take the color out too. Other than that, I can't think of anything -- sorry.

Nail polish remover (acetone) will remove nail polish from your nails, so it should remove it from anything else too, but try it in an inconspicuous spot. I have a feeling acetone will melt polyester, so if that's the material that is stained, be careful.

Yes! If its on a counter top or some sort of hard surface, you can use nail polish remover to rub it off. Be gentle and make sure you don't rub off the surface itself. I would advise you use cotton balls, dip it half way into the remover, and scrub softly.

well, what is it on?.. you have to give us a little bit more detail..

but, as far as fabrics and hard surfaces, its time to call upon the trusty nail polish remover. though it may smell a little bit, you may want to wash it afterwards.. if its a fresh stain (which i assume it isn't..) you could quickly wipe it off with a little water.
