Yes, new spyware and viruses are being developed all the time so there's always a chance something could slip through the net. However keeping your antivirus and anti-spyware up-to-date is the best defence available. Many would also suggest using Firefox as your browser rather than IE to reduce the risk of spyware.
Although I'm not a huge Microsoft fan in terms of their security, I have found Windows Defender to be a very good, free, easy to use anti-spyware product.

As new viruses and spywares are being created daily, there is always a change the lastest one may get through. That's why you have to keep your programs up to date. Most anti-whatever programs check for updates either daily or everytime you run them. Plus you should make a regular habit of running a full system scan regularly. (How often depends on how much time you spend on the Internet or a network.)
Plus, the list of what is considered at virus or a spyware are maintained by human beings, so they are only as accurate as the people who maintain the list. That is why I always recommending to TWO anti-spyware programs on your PC. One may catch something the other misses. (You can only run one anti-virus program at a time.)

There are an average of 2 dozen new malwares that come out each week. NO Safety program can guarantee 100% safety, but the good ones (free or pay) try there best to keep up.
What ever you use it's VERY important to keep them updated.
The MOST affective way to keep safe is to practice safe computing. It's like having a 3-inch thick steel front door, but then you let the crook with the gun in yourself! No amount of protection can protect your computer from you.
I use all of the following Free software with Windows XP SP2 and Internet Explorer with MSN toolbar and the pop-up blocker turned on and have never had a single problem with my computer sense Microsoft came out with SP2. (knock on wood.) (And be sure to keep Windows and the following software Updated)
The best Anti-virus is: AVG Anti-Virus Free Edition. (www.grisoft.com)
The Best Spyware defense is: SpyBot Search and Destroy (http://www.safer-networking.org/en/index.html)<--English version
AND: AdAware SE (www.lavasoft.com)
AND Spyware Blaster (http://www.javacoolsoftware.info/kb/)
The best Firewall besides the one that comes with Windows is: Zone Alarm (http://www.zonelabs.com/store/content/co... or just (www.zonelabs.com) and select the FREE version.
These are all FREE and are all that are required. There is no reason to pay for this kind of software, except to give a donation to the writers if you can.
ALL the above programs are recommended and should be run on a regular basis. With some the free versions they must be run manually.
The best way to avoid getting any Malware (virus, Trojan's, spyware, etc) is to practice safe computing. Microsoft and many others have countless articles on the subject of safe computing. (something I think should be taught in schools)
