Right-click "My Computer", click "Manage", click "Disk Defragmenter" under "Storage" in the left pane, click the "C:" drive on the right (or whatever drive you wish to defrag) and click the button "Defragment". You can schedule this as well.
Diskeeper is pretty good commercial software that you can use as well if you're nuts about wanting to keep your drive defragmented because it will do this "on the fly."
Otherwise, there's a free utility called "SpeeDefrag" that you can download and install from http://www.vcsoftwares.com/SpeeDefrag.html. This will help keep things easy for you.

if you have windows hit the start button .go to all programs or find accessories then you will see system tools open that up then u will see defrag click defrag and you r ready 2 go.also you should do a disk clean up.i defrag every 4 or 5 months just to keep my system in order.

go into safe mode then follow smitty's advice. Do this before you go to bed and with any luck it will be finished by the time you wake up, depending on the size of the drive

just in case you have linux, don't worry about it...
linux boxes don't need to be defragged...(at least, not if you're using ext* filesystems)
