YES YES YES If you spill just enough to get the padding wet and it doesn't dry it will mold. Then will spread. Hook up a fan and see if that will help. It should.

Yes,spills and spots need immediate attention. A liquid spill, even if it is only water, will act as a magnet to any dirt particles that are in the rug, causing the dirt to rise to the surface. This dirt may cause a dark stain. To prevent this from happening, try to keep the rug as soil-free as possible by daily vacuuming.
When a spill occurs:
BLOT , do not rub, the area with a clean, absorbent white towel. Never rub the area of the spill. Rubbing can work the liquid deeper into the fibers, making the spill more likely to stain the carpet. It can also spread the liquid to a larger area of the rug.

Not really. The water in the carpet will evaporate in minutes.
Maybe if you lived in an extremely dark, damp house.

If we're talking a pitcher full of water...not really. It should disperse quickly enough to not become a problem.
If we're talking 30-40 gallons like any aquarium full of water... yes, there's enough to make an impact.
