In modern times, I think Einstein's Theory of Relativity. It gave us a great understanding of how space, time gravity and light relate to each other,and the origins of the Universe, opening our minds to other great discoveries like quantum mechanics and putting us in route for a theory that would unify who the entire Universe works.

Maybe is was the discovery or creation of Calculus by Newton. Without the appropriate language it's hard to move forward.
