Every single one of them should be wheelchair accessible.
The Adams Mark is very nice, expensive, but right on the beach.
But frankly, if you can, I would go further north. St. Augustine Beach is way better than Daytona. The beach is wider, you can drive on it just like Daytona, and it's a lot less crowded. Same with Crescent Beach, just south of St. Augustine.
A day trip into St. Augustine is well worth it. There's a ton of history there, and a million little shops for the wife. It's where my husband and I honeymooned. Also the ghost tour at nighttime is highly reccommended.

If you do stay in Daytona, I suggest dinner at Auntie Catfish- by the Dunlawton Bridge.

Apparently, there are 43 wheelchair accessible hotels in Daytona Beach... check out the link below. It just comes down to how much you want to pay and what amenities you want for your wife.

Hi, they show several hotels on those sites, on the second one you can check online for room availability. Daytona Beach is really wonderful. Have fun with your wife.

Most hotel chains now required their hotels to have Handicapped Accessible facilities. Try something with a brand name you know. Best Western, Comfort Inn, Quality Inn, Radisson, Marriott. If you are unsure about their H.A. status, give the property a call.

I believe that they all should accomodate you, I would try expedia.com or travelocity for reputable information and even discounted rates that they don't advertise. Good luck and have a great trip!!
PS: Avoid Marriott properties
