Presumably you already know _how_ to edit the registry (regedit.exe, or programmatically) - but have found that Windows won't allow it. If this is the case, then the chances are you're on a network and have had your rights limited by the administrator for a perfectly valid reason. Overriding this and effectively "hacking" your workstation is going to REALLY annoy the administrator - I wouldn't recommend it!

Well, you need to use an account with administrative rights to edit the registry. To access Windows registry editor go to Start menu > Run > type "regedit" (without the quotation marks) and [Enter]. A new explorer window will open. You can now make edits to the registry.
