To answer this question, you need a Figure of Merit, something that reflects what is important to you in a car. Manufacturers typically quote mpg, 0-60mph acceleration time, OTR price, engine capacity, top speed, power-to-weight ratio. You can use these singly or in combination (e.g with weightings) according to your preference. Then you apply the Figure of Merit to a range of vehicles, allowing you to compare them consistently. A few things to consider:
you could use the square of the top speed to give fast cars that added edge
you could have an objective measure of desirability, according to marque
you could include perceived reliability
Here's an example:
Figure of Merit = power-to-weight ratio (kW/kg) + (top speed)^2 + desirability
Another example:
Figure of Merit = (1/weight) + (mpg)^2

No.
If it was, they wouldn't have replaced it.
They introduced the Murcielago because they believed (and I've yet to read a report that says otherwise) that it was better than the Diablo.

No. The best cars in the world are made by Aston Martin. They outstrip all others and there is no contest.

You can't really call any car the "best" car but it could be your favorite. My personal favorite is the buggati veyron (it is also the fastest if that is what your asking).
