generally how would you set it up, common problems that occour and how you would go to fix them.
generally compnays will not wants scripts syntactally 100% spot on (but it helps), but that you know how to go about as some people do get interview jitters and make a slight mistake on syntax, but if they can tell you why this wouldnt work n y and what your would do to fix it.
sort of questions i have been asked are :
What is an Mx record.
What are valid values for an Mx record? (IP, Glue, Cname?)
What happens if two mx records have the same preference? Different preference?
What is a TTL value? And why should I care?
What is the difference between EHLO and HELO?
Describe the IIS SMTP service.
Others might be more concerned about architecture:
Describe how you'd configure a server for $number users.
Which is better? More databases in fewer storage groups or more storage groups with fewer databases? Why?
Others might be more concerned about day to day user support:
Jane has gotten married, how would you modify her account?
The CEO can't connect to Exchange, he's getting the error 'unable to open your default folder', what would you do to troubleshoot?

Having had several of these thrown at me in the past, I can tell you that usually they are asked as a result of a somewhat recent bad experience by the company that they struggled to resolve. As such, you'd be best to get a little intel info on what's been going on with them before hand if you aren't comfortable with your level of knowledge.
Of course if you know the system, you can probably reason through most issues.
