IMHO, the title is held today by two: Melissa Etheridge and Elton John.
These two have done more for positive public relations in the Straight world for the Gay community by writing and singing their songs.
Ellen DeGeneres and Rosie O'Donnell run a very close second for their television shows.
There are countless other Gay folks who have done unknown work behind the lines for Gay human rights advocacy. Sadly, they will never come into the limelight of fame and be acknowledged for their valuable contributions. But they deserve to be considered Icons of honorable mention none the less.
PS: The founder of Stonewall is Sylvia Revera, the famous Transwoman.

In no particular order here are some of these might qualify, not all admittedly:
Oscar Wilde, James Dean, Rock Hudson, MIchaelangelo, Leonardo Da Vinci, Tennessee Williams, Pasolini, Jean Cocteau, Tom of Finland, David Hockney, Andy Warhol, Joe Dallesandro, Kenneth Williams, Quentin Crisp of course, Freddy Mercury..the list could go on and on I guess, I might come back when I think of lots more!
If we are talking acting & singing diva's:
Maria Callas, Shirley Bassey, Judy Garland, Bette Davis, Joan Crawford, Barbara Stanwyck, Barbra Streisand (in her early years) Liza Minella (as Sally Bowles in Cabaret), Madonna, Kylie Minogue, Boy George & Marc Almond not forgetting Jimmy Somerville, David Bowie & Lou Reed.

Madonna seems to be iconic in the gay crowd, not only for music but for her frank opinions and demonstrations of sexuality in many forms.

Im not gay, or pay attention to that community, but I think Ellen Degeneras is a groovy lady. I think Melissa Etheridge is cool too, but she's not as much a gay icon, in my mind, as Ellen is.

I would say Elton John, Cher, Madonna, Melissa Etheridge, and even Boy George. These are singers who are famous and very open in the community.

I had the unique pleasure of speaking with this man. He was unique, fearless, uncompromising and a true Free Spirit who, to me, was the singular most important Gay figure to bring awareness of the plight of the GLBT community on a global scale: QUENTIN CRISP.
Best Wishes!
