There are many but many within the early years of the many of the important court rulings changed the structure of government and set the direction for the future.

A few are:

Marbury v. Madison [1803] Began the defining of a foundation for the separation powers as defined by the Constitution and was elemental in the establishment of “Judicial Review.”.

Dartmouth College v. Woodward [1819] Court ruled that a charter of incorporation was a contract protected against legislative infringement.

McCulloch v. Maryland [1819] In this decision the court referred to the 18th Clause of the 8th Section of the 1st Article, stating that the words “necessary” and “proper” had the same meaning. This changed the power of the Congress to enact laws outside of its delegated powers.

Marbury vs. Madison (1803): Established the Court's right to review the constitutionality of governmental actions. Arguably its most important case.

Brown vs. Board of Education of Topeka, Kansas (1954): Struck down its previous ruling in Plessy v. Ferguson: declared segregation unconstitutional.

Roe vs. Wade (1973): Established privacy as an implied right in the Constitution. Applied that privacy right to abortion in certain situations. Held that plantiffs must sbe seeking relief from an actual case or controversy -- essentially, they must demonstrate that they are harmed, not that they may be at some point in the future.
