Many people think that the only difference is the DH, but they are wrong. Now I can't name off hand the other subtle differences but they have to do with substitutions and other things like that. Managing in the AL is much different than the NL.

The Designated Hitter: another player bats instead of the pitcher. It's used in the American League, but not the National League.

The major difference is that the American League has the Designated Hitter to bat instead of the pitcher, where as in the National League, the pitcher bats.

Pitcher bats in National league while there is a designated hitter, which is a guy who only bats and doesn't field, in the American league. As a result, there is more pinch-hitting, hit-and-run, and more bench use in the National league.

First, American League has a bater that only bats and doesn't play defensively..... This batter is called the designated hitter. National League doesn't have a designated hitter, instead the pitcher bats.

American league has a designated hitter and National league is when they don't have a designated hitter and have the pitchers hit.

haha so in lame terms, nothing. they just have it split in two leagues to make life easier. though the pitcher bats on the national side and the american side has a DH is beyond me. i guess to give pitchers a choice of either A) wasting their power by pitching and batting or B) save ur power and make someone else do it.
