Lord of the Rings, Return of the King. It won 11 Academy Awards without a single acting award. Had it won an acting award, it would be (and should be) the most honored film in history surpassing Ben Hur and Titanic.
This movie took 7 years to bring to the screen. New Line Cinema put the future of their entire company in the hands of a director who never had a big hit previously and he (Peter Jackson) delivered a film - a trilogy, really - that is at once visually stunning, emotionally gripping and totally rewarding to the viewer.
My 2nd favorite is Pulp Fiction which I feel is the Best original screenplay ever - and has some all time memorable lines.

Its almost impossible to compare one genre to another, but if I had to pick one movie I'd go with THE WIZARD OF OZ.
THE GODFATHER, TO KILL A MOCKINGBIRD, PSYCHO, and CASA BLANCA would be in consideration as well.
