"Hot"... add it to the tank to the line marked hot.
Use the recommended mix i.e. 50/50 water antifreeze.
It may be a good time to add "corrosion inhibitor", that is cheap, and some "water wetter" it will increase the cooling capacity of your system.
Both are available at the auto-parts store.

If your cooling system is cold, you should fill to the radiator cap. Make sure to use a 50/50 mixture of antifreeze/coolant and water. You can also add a bit to the overflow tank. When your engine heats up, the coolant will expand and overflow into the tank. When your engine cools, it draws the overflow fluid back into the radiator. You don't want to overfill the overflow tank for that reason.

the fluid is added to the expansion tank on a regular refill but if it is done for the first time the addition goes into the radiator only

Radiator itself although I don't think any harm can happen if you used the expansion tank.
One drawback of the expansion tank is that it usually is the rusty old, dirty anti-freeze. Your just dluting dirt.
Adding it to the radiator, you can drain the old out and flush the radiator to remove debris from the wingnut located at bottom of radiator.
